    (I) Purpose
    Vesicle Prep Pro (VPP) is a product of Nanion, Germany. It is the world's first fully automatic equipment for preparing giant unilamellar vesicles (GUV). It mainly uses electro-swelling technology (i.e., hydrating dry lipids under an alternating electric field) to quickly prepare vesicles. The diameter of the vesicles ranges from 1-30 μm, and they have the characteristics of strong stability and good uniformity. They are mainly used in lipid bilayer experiments to study the biophysical properties of some substances, such as lipid membranes, electrically active transmembrane protein channels, lipid rafts, and lipid-mediated DNA transfer, etc.

    (II) Instrument composition
    The vesicle preparation instrument mainly includes the vesicle preparation instrument host, vesicle preparation tank, indium tin oxide glass slide (ITO glass slide) and VesicleControl software. It has two control modes: 1. Manual control: the parameters of vesicle preparation can be set directly by adjusting the knob on the instrument host to prepare vesicles. 2. Software control: the parameters of vesicle preparation can be set through VesicleControl software to prepare vesicles. It is recommended to use the simpler manual control to prepare vesicles.

    Vesicle preparation instrument host: set various parameters for vesicle preparation, such as voltage amplitude (Ampl), frequency (Freq), rise time (Rise t), preparation time (Main t), fall time (Fall t), and temperature (Temp).

    Vesicle preparation tank: It fixes the ITO glass slide and the components for preparing vesicles. It has a heating element inside, and the temperature can be controlled by the host.

    Indium tin oxide glass slide: also known as ITO glass slide, one side of which is electroplated with a layer of indium tin oxide conductive material, and the other side is non-conductive.

     

    Features:

    Easy operation and fast preparation: According to the set program, the instrument automatically prepares stable and uniform vesicles. The preparation time is 2-4 hours (adjusted according to the type of phospholipids used to prepare the vesicles).

    Temperature control: There are heating elements in the main unit and vesicle preparation tank, which can adjust the temperature as needed.

    Visualization: The vesicle preparation tank can be placed under a microscope to observe the vesicle preparation process in real time.

    Strong repeatability: The formation of vesicles is controlled by program, and the preparation process is programmed, reducing the impact of more human operations.

    Good uniformity: Most of the prepared vesicles have similar diameters and small size differences.

    Good stability: The vesicles can be stored in a refrigerator at 4°C for up to 4 weeks. The lipid bilayer membrane is stable when recording lipid bilayer experiments.

     

    Product Application:

    The vesicle preparation instrument is an instrument for preparing uniform and stable artificial lipid vesicles. The vesicles can be recombined with channel proteins to form liposomes, which are used in lipid bilayer experiments.

     

    Parameter function:

    Freq: The frequency of the electric field change.
    Ampl: voltage amplitude of vesicle preparation.
    Rise t: The time required for Freq and Ampl values to reach the set values from 0.
    Main t: time for preparing vesicles.
    Fall t: The time required for Freq and Ampl values to drop from the set value to 0.
    Temp: The set value of temperature during the preparation process.
    The voltage amplitude, frequency and preparation time directly affect the size and uniformity of the vesicles. The standard protocol is: frequency 5Hz, amplitude 3V, preparation time 120 minutes, rise time 3 minutes, fall time 5 minutes, temperature 36℃.
